Enzyme kinetic analysis reveals a dynamic relationship between enzymes and their substrates. Overall enzyme activity can be controlled by both protein expression and various cellular regulatory systems. Interestingly, the availability and concentrations of intracellular substrates can constantly change, depending on conditions and cell types. Here, we review previously reported enzyme kinetic parameters of cellular and viral DNA and RNA polymerases with respect to cellular levels of their nucleotide substrates. This broad perspective exposes a remarkable co-evolution scenario of DNA polymerase enzyme kinetics with dNTP levels that can vastly change, depending on cell proliferation profiles. Similarly, RNA polymerases display much higher Km values than DNA polymerases, possibly due to millimolar range rNTP concentrations found in cells (compared with micromolar range dNTP levels). Polymerases are commonly targeted by nucleotide analog inhibitors for the treatments of various human diseases, such as cancers and viral pathogens. Because these inhibitors compete against natural cellular nucleotides, the efficacy of each inhibitor can be affected by varying cellular nucleotide levels in their target cells. Overall, both kinetic discrepancy between DNA and RNA polymerases and cellular concentration discrepancy between dNTPs and rNTPs present pharmacological and mechanistic considerations for therapeutic discovery.The evolutionarily conserved multiprotein Mediator complex (MED) serves as an interface between DNA-bound transcription factors (TFs) and the RNA Pol II machinery. It has been proposed that each TF interacts with a dedicated MED subunit to induce specific transcriptional responses. But are these binary partnerships sufficient to mediate TF functions? We have previously established that the Med1 Mediator subunit serves as a cofactor of GATA TFs in Drosophila, as shown in mammals. Here, we observe mutant phenotype similarities between another subunit, Med19, and the Drosophila GATA TF Pannier (Pnr), suggesting functional interaction. We further show that Med19 physically interacts with the Drosophila GATA TFs, Pnr and Serpent (Srp), in vivo and in vitro through their conserved C-zinc finger domains. Moreover, Med19 loss of function experiments in vivo or in cellulo indicate that it is required for Pnr- and Srp-dependent gene expression, suggesting general GATA cofactor functions. Interestingly, Med19 but not Med1 is critical for the regulation of all tested GATA target genes, implying shared or differential use of MED subunits by GATAs depending on the target gene. Lastly, we show a direct interaction between Med19 and Med1 by GST pulldown experiments indicating privileged contacts between these two subunits of the MED middle module. Together, these findings identify Med19/Med1 as a composite GATA TF interface and suggest that binary MED subunit-TF partnerships are probably oversimplified models. We propose several mechanisms to account for the transcriptional regulation of GATA-targeted genes.
The Person-centered Care Assessment Tool (P-CAT) was developed as a self-reporting assessment scale for the healthcare staff ratings of the person-centeredness of their nursing practice.
This study investigates the psychometric proprieties of P-CAT tool in a sample of staff working in residential units for older people, in the North of Italy.
Internal consistency and reliability were examined using the Cronbach’s alpha coefficient. Exploratory factor analysis was used to evaluate construct validity, homogeneity analysis performed to evaluate internal homogeneity of the items and equidistance of item options, test-retest reliability examined by the Pearson correlation coefficient and the intraclass correlation (ICC) coefficient. The P-CAT score was standardized to a 100-point scale, the score differences among groups were compared with one-way ANOVA.
The exploratory factor analysis supported the construct validity of a two-factor solution. The mean standardized score of P-CAT was 67.3 (
12.8) and Cronbach’s alpha was .79 for subscale 1 and .75 for subscale 2. The ICC coefficient was .87.

Childbirth can have negative consequences; however, measurement tools to assess this event are limited; and none consider the adolescent’s birth. This study assessed the psychometric properties of the childbirth trauma index (CTI).
Construct validity and reliability of the CTI was assessed via a field test involving 160 adolescents 3 days postpartum.
A modified 8-item version of the original CTI exhibited acceptable construct validity and reliability criteria. The CTI was found to link with birth appraisal, but not subjective distress measurements.
The CTI may be a more robust means of assessing birth appraisal than use of single-item measures. Recommendations for practice suggest use of the modified CTI to assess birth appraisal, or use in combination with single-item rating scales.
