These results suggest that the ocean’s twilight zone receives a consistent diurnal supply of labile DOM from the diel vertical migration of fishes, enabling an unexpectedly active community of heterotrophic prokaryotes.Pteris cretica var. nervosa is a dominant fern species found in antimony (Sb) mining areas, capable of forming symbiosis with arbuscular mycorrhizal fungi (AMF), especially with those members of the Glomus genus. Despite this fern’s relevance and the potential contribution of mycorrhizal symbiosis to phytoremediation, the AMF’s impact on P. var. nervosa phytoremediation of Sb remains unknown. Here, we exposed P. var. nervosa to different concentrations of Sb for 6 months. Our results showed that Sb reduced shoot biomass, enlarged the root/shoot ratio, and disrupted the fronds’ intracellular structure. AMF inoculation, however, was able to moderate these phenotypic changes and increased the accumulation level of Sb in plants. From a proteomics analysis of this plant’s fronds, a total of 283 proteins were identified. Notably, those proteins with catalytic function, carbon fixing and ATP metabolic function were highly enriched. K-means clustering demonstrated protein-changing patterns involved in multiple metabolic pathways during exposure to Sb. Further, these patterns can be moderated by AMF inoculation. Pearson correlations were used to assess the plant biomarkers-soil Sb relationships; This revealed a strong correlation between ribosome alteration and the root/shoot ratio when inoculated with AMF, and a positive correlation between photosynthesis proteins and chlorophyll (SPAD value). Our results indicate AMF could moderate the fronds impairment by maintaining the sufficient protein levels for ribosomal functioning, photosynthesis activity and to counter ROS production. Wuzhi capsule (WZC) is a traditional Chinese medicine preparation; it is used to decrease the aminotransferase level of the liver and protect liver function. Thus, patients with hepatocellular carcinoma (HCC) are potentially treated with a combination of LEN and WZC, but there is no information about the interaction between the two drugs. We developed a simple, rapid, and sensitive ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method for the quantitative determination of lenvatinib in rat plasma. Liquid-liquid extraction of plasma samples was carried out with ethyl acetate. Chromatographic separation of analyte was performed using gradient elution with acetonitrile and 0.1% formic acid water. The positive ion multi-response monitoring mode was used, and the target of the parent and daughter ions of LEN and IS were m/z 427.1→370 and m/z 432.1→370, respectively. All the validation projects were in accordance with the guidelines. Good linearity of 0.2-1000 ng/mL (r > 0.999) was achieved. The lower limit of quantification was 0.2 ng/mL. The methods were successfully validated based on internationally accepted guidelines (EMA, FDA), encompassing selectivity, evaluation of the calibration model, matrix effect and recovery, accuracy and precision, stability, and dilution integrity. In addition, for the VAMS samples, the effect of the hematocrit on the recovery was evaluated. Successful application on whole blood and plasma, as well as on VAMS samples prepared from venous or capillary blood of patients, demonstrated that the methods were fit-for-purpose and can be used for future PK studies.
Despite research linking dissociation, alexithymia, and anger with childhood trauma and ADs, the investigation addressing the relationships between the potential mediators has not yet been established within the literature.
The present study examined the relationship between childhood trauma and autoimmune disorders, using a multiple mediation model that included dissociation, alexithymia, and anger as hypothesized mediators.
306 autoimmune patients and 292 self-declared healthy controls were included in the study. They completed self-report questionnaires regarding childhood abuse, alexithymia, dissociation, and anger.
Multiple mediation analysis was conducted to investigate the study’s proposed model.
The results of the Structural Equation Model (SEM) suggest an indirect relationship between childhood trauma and autoimmune disorders, mediated by dissociation [z=4.57, p<.01, β=0.19, 90% CI (0.08-0.10)], alexithymia [z=10.74, p<.01, β=0.43, 90% CI (0.08-0.10)], but not by anger [z=1.58, p=.11, 90% CI (0.08-0.10)].
These findings are in line with previous studies that show associations between childhood trauma, dissociation, alexithymia, and ADs. They indicate that mental health professionals and medical doctors should assess childhood trauma in autoimmune patients. They also should consider the possible maintaining role of dissociation and alexithymia in the treatment of autoimmune diseases.
